چکیده

Intra nasal drug delivery system (INDDS) has emerged as a noninvasive alternative for delivering peptides, proteins and other therapeutic agents, specifically to CNS (Central nervous system). This route is well suitable for systemic delivery because of high vasculature, permeability, elimination of hepatic first pass metabolism and low enzymatic environment of the nasal cavity. This route of delivery is especially suited to treat CNS disorders like neurodegenerative diseases, mood disorders, cognitive dysfunctioning etc., due to drugs ability to directly reach the brain by passing blood brain barrier (BBB). It is also a well suited delivery route for the biotechnological products like DNA plasmids, hormone, proteins, peptides, DNA vaccines to give enhanced bioavailability. A wide variety of therapeutic compounds may be administered intra-nasally for topical, systemic and CNS disorders. In this review, we have focussed on the nasal passage anatomical pathway leading to CNS drug delivery, different factors affecting nasal absorption, bioavailability barriers, vaccines and other therapeutic, biologically active molecules delivered via intra nasal route and strategies to improve nasal absorption. We have also outlined various in vitro and in vivo cellular models for nasal drug absorption and permeability studies. INTRODUCTION: Intra nasal drug delivery (INDD) system was used for intake of various medicinal compounds in Indian Ayurvedic and yogic medicinal practices by referring them as ―Nasya karma‖ or ―Nasya rasayana‖ from last thousands of years. And in today’s modern medicine concept also, in response to the insufficiency of conventional delivery mechanisms, nasal drug delivery (NDD) route has been extensively explored as an alternative system to promote more effective delivery of drug molecules 1, 2 .

Freeze-dried k-carrageenan/chitosan polyelectrolyte complex-based insert: a novel intranasal delivery system for sumatriptan succinate

Intranasal route, ensuring suitable bioavailability of medicines under circumvention of the gastrointestinal degradation and hepatic first-pass elimination, has been a popular choice for drug delivery. Among nasal dosage forms, mucoadhesive solid inserts have been shown to resist mucociliary clearance and provide a prolonged nasal residence time. Hence, the purpose of this study was the prepara...

Lanthanides are a family of elements, commonly known as rare earth elements. Metal samarium as one of the most important member of this family, which plays very important roles in the diagnosis and treatment of various diseases, especially cancer. Several lanthanide complexes, samarium in particular, have shown anticancer effects on tumor cells. This paper is tried to review the biological prop...

The nose may help the brain: intranasal drug delivery for treating neurological diseases

While enormous progress has been made regarding our understanding of the pathogenic mechanisms of neurological diseases, there are only a small number of effective drugs for treating these illnesses. A key obstacle for developing effective drugs for treating neurological diseases is the blockage of drug entrance into the CNS by the BBB [1]. Less than 2% of all small-molecule drugs, and virtuall...

Intranasal Oncolytic Virotherapy with CXCR4-Enhanced Stem Cells Extends Survival in Mouse Model of Glioma

The challenges to effective drug delivery to brain tumors are twofold: (1) there is a lack of non-invasive methods of local delivery and (2) the blood-brain barrier limits systemic delivery. Intranasal delivery of therapeutics to the brain overcomes both challenges. In mouse model of malignant glioma, we observed that a small fraction of intranasally delivered neural stem cells (NSCs) can migra...
